中文摘要: 生物体生命活动的绝大部分能量来自于呼吸,对生物呼吸作用全局调控模式的研究有助于加深对生命生存和环境适应基本机制的理解。深海细菌通常能利用多种末端电子受体进行厌氧呼吸,具有呼吸多样性。对深海细菌厌氧呼吸机制的研究不仅是理解深海细菌极端环境适应机制的关键,还有助于对地球早期生命的生存方式及其演化的探索和理解。本研究以分离自西太平洋深海沉积物中的嗜冷耐压细菌Shewanella piezotolerans WP3为研究对象,结合比较基因组学,转录组学等生物信息学分析和基因敲除,功能验证等各种分子生物学技术,对WP3呼吸系统(特别是厌氧呼吸)的组成、调控因子的转录表达和呼吸作用的全局调控进行系统分析。本研究将是揭示深海细菌环境适应调控机制和代谢演化的重要基础。

英文摘要: Respiration is required for living organisms to obtain enough energy, thus studies on global regulation of respiration will improve our understanding of basic mechanisms of survival and adaptation to environment. Deep-sea bacteria are known for their diversities in respiration and usually able to use a variety of terminal electron acceptors for anaerobic respiration. Studies on regulatory mechanisms of anaerobic respiration not only are of great importance to understand adaptations of deep-sea bacteria to extreme environments, but also contribute to explorations of early lives on Earth and understandings on life forms and evolution. In this study, Shewanella piezotolerans WP3 separated from deep-sea sediments in Western Pacific is used. With techniques in comparative genomics and transcriptomics analysis, bioinformatics and molecular biology, composition of the respiratory system (in particular anaerobic respiration), expression profiles of regulatory factors and global regulation of respiration will be studied in a systematic way. This study will be of great importance for uncovering mechanisms of adaptation and metabolic evolution of deep-sea bacteria.
