中文摘要: 本项目研究类肝素结构天然生物大分子水凝胶的合成及其在血液接触材料和吸附分离领域的应用。设计合成出同时含有磺酸基和羧基的类肝素结构壳聚糖(HLCS),再将HLCS制备成水凝胶,降低其在血液接触时对血浆蛋白的吸附,延长凝血时间,抑制凝血途径的激活和补体的激活。研究磺化剂/羧化试剂用量、反应时间和反应温度等对HLCS中磺酸基和羧基接枝量及其比例的影响。系统考察磺酸基、羧基及其比例对水凝胶抗凝血性、pH敏感性、重金属离子(Cu2+等)和阳离子染料(亚甲基蓝MB等)清除率的影响。通过类肝素生物大分子制备水凝胶,解决了天然高分子水凝胶的抗凝血性能,赋予类肝素CS水凝胶良好的pH敏感性,制备的类肝素CS水凝胶同时在血液接触材料及吸附分离方面有很大的潜在应用。

英文摘要: In this project, a series of heparin-like chitosan (HLCS) are synthesized via two-step reactions, and then chemical cross-linking of HLCS results in the corresponding hydrogels. HLCS hydrogels could reduce the protein adsorption, prolong the clotting times, and prevent the contacting activation and complement activation. The influence of the amount of sulfonating agent and carboxylation agent, reaction time and temperature on the degree of sulfonation and carboxylation of HLCS will be investigated. The project will systematically study the effect of the degree of sulfonation and carboxylation of HLCS on the anti-coagulant property, pH sensitivity, heavy metal ions and cationic dyes removal ability of HLCS hydrogels. The proposed method herein has solved the problem of the poor anticoagulant property of polymeric hydrogels, and supplied good pH sensitivity for these polymeric hydrogels. These HLCS hydrogels have great potential to be used in the fields of both blood-contacting materials and adsorption-separation technology.
