中文摘要: 慢性肾脏病(Chronic Kidney Disease, CKD)因其高发病率、可进展至肾衰竭、易并发严重心血管疾病及认知障碍而成为全球性公共健康难题。近年来,CKD终末期肾病阶段脑功能及结构改变机制受到认知神经科学、肾病科学及临床应用的极大关注,但对CKD其它阶段相应的研究较少;尽管多模态影像技术已用于多种疾病的脑功能-结构-行为关系的研究中,但其在CKD各阶段大脑功能及结构改变研究中的运用却非常有限。据此,本项目拟采集CKD大样本的多模态脑影像结合数据分析,联合神经认知量表及临床生化指标,探测CKD不同阶段的脑局部特性、全脑网络特性的动态变化;另外,采用模式识别的方法,揭示CKD进展过程中大脑改变的早期影像学证据。本研究可望在理论上揭示CKD进展中的脑改变机制,获得其大脑改变的早期影像学证据。在临床上可预防或延缓CKD患者大脑不可逆性损伤、降低CKD认知障碍的发生率及其死亡率。

英文摘要: Chronic Kidney Disease (CKD), as its high incidence, can progress to kidney failure and easily be complicated with by severe cardiovascular disease and cognitive impairment, has become a global public health problem. Recently, investigations on the brain functional and structural change mechanism of ESRD patients have been intensively discussed in cognitive neuroscience, nephrology science and clinical application. However, the corresponding researches of other stages of CKD are relatively less. Despite the multimodal magnetic resonance imaging (MRI) has been used in analysing the relationships between brain function-structure-behavior and various diseases, the studies which observe brain function and structural changes about large other stages of CKD patients are very limited. Accordingly, this project will collect multimodal brain MRI data of large sample CKD patients combined with imaging data analysis, refer to the testing results of neurocognitive scales and clinical biochemical parameters, detect the local and global dynamic changes of brain network characteristics about different stages of CKD patients. In addition, this project, which using multiple voxel pattern analysis method, can reveal early neuroimaging markers of brain changes in the progression of CKD. In theory, this project can reveal the brain changes mechanism and provide early neuroimaging markers in the progression of CKD. The findings of this project can be adopted to prevent or delay the irreversible brain damage in patients with CKD and reduce the morbidity and mortality of cognitive impairment clinically.
