中文摘要: 高压静电场生物效应一直是国内外交叉学科领域的研究热点之一,目前关于高压静电场(HVEF)对生物体物理损伤机制的研究未见报道,HVEF生物效应的研究只在细胞组织水平上,更深入的分子水平的研究相对滞后。申请者在前期研究中发现高压静电场(HVEF)可以使大肠杆菌(E.coli)K12 W3110的诱变率较对照组提高10倍以上;本项目拟以该E.coli的lacI基因作为突变谱研究的目的基因,研究HVEF作用下E.coli的存活率、突变率及其导致的突变谱,从而获得HVEF导致遗传变异的分子机制;采用微生物细胞荧光标记与流式细胞仪(FCM)结合技术,对HVEF处理的E.coli进行实时检测,考察其对E.coli膜的损伤、胞内酶活力的影响及膜损伤的可修复性,并辅以扫描电子显微镜(SEM)观察其对E.coli形态的影响,揭示HVEF对E.coli损伤的物理机制。

英文摘要: In recent years,the biological effects of high voltage electrostatic field(HVEF) are worldwide research hotspots in the field of cross disciplines.But no reports are found in the physical damage mechanism of HVEF on organism,and the biological effects of HVEF had been studied broadly at the cellular and tissue levels,several researches were done at the molecular levels.We found that the HVEF can improve the mutational frequency of E.coli about 10 times more than contral groups.In this project,the survival rate of E.coli,the mutational frequency and the mutational spectrums of the lacI gene from E.coli by HVEF treating will be studied,from which the mutational mechanism of HVEF will be obtained.And the damage effects of HVEF on E.coli cell will be measured in real time by using flow cytometry(FCM) in combination with fluorescent techniques, including the damage of cell membrane,the change of endoenzyme activity and the reversibility of membrane damage.And the form of E.coli cell will be observed with the scanning electron microscope,at last the physical damage mechanism of HVEF to E.coli will be analysised.
