中文摘要: 马铃薯是我国第四大粮食作物,开展块茎发育的分子机理研究对于理解马铃薯产量控制机制具有重要理论意义。阿拉伯半乳糖蛋白(AGPs)是一类存在于高等植物中的羟脯氨酸富集糖蛋白,在茎的伸长和分支中起着调控作用。多种证据表明,马铃薯AGP基因可能参与控制块茎的形成和发育。本项目首先利用生物信息学方法从马铃薯基因组中鉴定获得116个StAGP基因,通过高通量表达分析筛选得到15个在匍匐茎和块茎中特异或高丰度表达的StAGP基因;拟采用基因表达模式分析、蛋白亚细胞定位、过量表达、基因沉默、基因编辑和蛋白纯化等多种分子生物学和生物化学手段,研究2-3个StAGP基因在块茎发育过程中的生物学功能;揭示StAGP基因控制块茎发育的分子生物学机理,建立StAGP基因参与激素信号传递的调控网络,探索StAGP糖生物学特征与蛋白功能之间的关联。本项目关于马铃薯块茎发育的分子机理研究将具有重要理论意义和潜在应用价值。

英文摘要: Potato (Solanum tuberosum) is the fourth crop in China, and studies on molecular mechanism of tuber development have important theoretical significance for understanding the mechanism of potato yield. Arabinogalactan proteins (AGPs) are a kind of hydroxyproline-rich glycoproteins that widely distributed in higher plants and implicated in regulating stem elongation and branching. A variety of evidences indicated that AGP genes might be involved in controlling potato tuber development. In this study, a total of 116 StAGP genes of potato was identified by using bioinformatics methods, and 15 StAGP genes that specifically or dominantly expressed in stolon and tuber were screened out based on high-throughput expression analyses. Moreover, multiple techniques of molecular biology and biochemistry will be utilized to investigate the biological functions of 2-3 StAGP genes, including gene expression pattern, protein subcellular localization, over-expression, gene silencing, gene editing, protein purification, and so on. For discovering the molecular mechanism of StAGP genes in controlling tuber development and building a regulatory network of StAGP genes in phytohormone-related signal transduction, our study will have important theoretical significance and potential application value.
