中文摘要: 本实验从隔药饼灸对血脂的调节作用及对血管内皮细胞具有保护作用的思路出发,采用隔药饼灸,以动脉粥样硬化(AS)模型兔为受试对象,运用生化放免、免疫组化、电镜技术和荧光定量PCR技术,比较隔药饼灸、直接灸对AS兔血脂、主动脉内皮细胞TLR4抗原免疫活性、TLR4mRNA表达及主动脉内皮细胞超微结构的变化情况,从不同角度、不同水平对隔药饼灸在动脉粥样硬化过程中的调控作用进行了实验观察,具体研究结果如下:①#38548;药饼灸能有效降低AS兔血中总胆固醇(TC)、甘油三酯(TG)含量,且能降低血中低密度脂蛋白(LDL)和载脂蛋白B(apo-B)含量,升高高密度脂蛋白(HDL)和载脂蛋白A(apo-A)含量,提示隔药饼灸对脂质及脂蛋白的代谢具有良好的调整作用。②#30005;镜技术显示隔药饼灸对AS兔主动脉内皮细胞有一定的保护作用。③#38548;药饼灸能抑制主动脉内皮细胞TLR4活性,抑制主动脉TLR4mRNA表达,提示隔药饼灸是通过抑制TLR4表达从而延缓AS的形成。④#26412;实验研究证实了隔药饼灸延迟AS形成的作用机制是通过阻断TLR4抑制信号通路后的炎症反应及免疫反应而实现的,为临床合理应用隔药饼灸防治AS提供了一定实验依据。

英文摘要: This research started from the thingking of herbal cake separated moxibustion 's regulating effect to blood fat and protection to vascular endothelial cells,chose herbal cake separated moxibution and took AS rabbits,used biochemical immune,immunohistochemical,electron technology and RT-PCR,compared the influence of herbal cake separated moxibustion and direct moxibustion to AS rabbits' blood fat,Ag activity of aortic endothelial cells'TLR4,the expression of TLR4 mRNA and the change of aortic endothelial cells' ultrastructures,observed herbal cake separated moxibustion's regulation to atherosclerosis process from different angles and levels.The definite research results:①erbal cake separated moxibustion can reduce AS rabbits' TC、TG、LDL 、apo-B effectually,escalate the content of HDL、apo-A,this promptes that herbal cake separated moxibustion has good accommodation to blood fat and lipoprotein metabolis.②ectron technology test shows herbal cake separated moxibustion has certain protection to AS rabbits'aortic endothelial cells.③erbal cake separated moxibustion can restrain the activity of aortic endothelial cells'TLR4 and restrain the exression of TLR4 mRNA.This indicates herbal cake separated moxibustion delays AS ' formation by restraining the exression of TLR4.④his research confirmed that the mechanism of herbal cake separated moxibustion delaying AS'formation is Blocking the inflammatory response after and immune response after TLR4'restraining signal pathway,provide some experimental basis for the reasonable application of herbal cake separated moxibustion Prevention and treatment to AS in clinical.
