中文摘要: 鄱阳湖是我国最大的淡水湖,日益面临各种潜在的重金属污染,镉(cadmium)是其中的一种典型有毒重金属。本项目选用鄱阳湖产草鱼为实验动物,建立草鱼镉中毒的试验模型,分离鱼体肝与肌肉细胞系进行体外培养。电镜下观察镉中毒后鱼体细胞内线粒体结构的变化,生化检测分析线粒体的功能变化及其对细胞凋亡相关基因表达的影响。从个体、细胞、亚细胞及分子水平,分别检测草鱼肝和肌肉细胞中线粒体的多种生物学指标,研究其变化与细胞凋亡之间的相关性,以验证线粒体在镉诱导草鱼肝和肌肉细胞凋亡中可能的介导作用,并探讨镉诱导草鱼肝与肌肉细胞凋亡的分子机理。本项目的完成,将为鄱阳湖水体重金属污染的生物监测提供资料,并为渔业生产安全评估与食品安全检测提供可靠依据。

英文摘要: As the largest fresh water lake in china, Poyang Lake is being threatened by various heavy metals pollution, including cadmium, a typical noxious heavy metal. In the present study, grass carp from Poyang Lake will be raised in water with cadmium to build an experimental model, with cell lines from fish liver and muscle separated and cultured in vitro, respectively. Structural changes within mitochondria of cadmium-challenged fish cells will be observed using electron microscope, and the effects of mitochondria's functional variation on expression of apoptosis related genes be detected and analyzed. Furthermore, various biological indexes of mitochondria of fish cells will be detected at individual, cellular, subcellular and molecular levels, respectively, and the relationship between its change and apoptosis will be studied to determine the potential mediated role of mitochondria during apoptosis induced by cadmium, and investigate its molecular mechanism deeply. Results of this study will provide not references for biological monitoring of heavy metal pollution of water in Poyang Lake, but basis for security evaluation of fishery production and safety measure of fish food.
