中文摘要: 病毒侵入寄主后会诱发各种病害症状,至今,对RNA病毒的病害症状形成的分子机制仍然知之甚少。本项目拟在前期扎实工作基础上,针对水稻草状矮化病分蘖增多的表型症状形成机制这一科学问题,采用Deep Sequencing技术、甲基化测序、生物信息学分析、基因芯片、Northern blot鉴定、转基因水稻验证、CO-IP等手段,研究RGSV 侵染后对水稻内源小RNA产生和作用的影响,明确RGSV调控水稻基因的甲基化机制及其过程,鉴定出在水稻基因甲基化过程中起关键作用的RGSV功能基因,探讨RGSV侵染后水稻分蘖增多的表型症状形成机理,这对深入揭示RNA病毒致病、植物病毒症状形成机制和寄主水稻抗病毒机理具有重要的理论价值,也为水稻草矮病的防控和抗病育种提供新策略、新思路。

英文摘要: Symptoms of virus-infected plants are often very specific to the given virus. The molecular mechanisms involved in viral symptom induction have been extensively studied, but are still poorly understood, especially for RNA viruses. This research is planned on the basis of preliminary work in the scientific issue: the formation mechanism of increased tillering symptom caused by Rice grassy stunt virus. We will use solexa deep sequencing, DNA bisulfite sequencing, bioinformation, microarray, northern blot, transgenic methods and CO-IP to illustrate the influence of RGSV in rice RNAi & RdDM pathway regulation, DNA methylation and its mediated disease symptoms. This work can help us reveal the pathogenesis of RGSV, as well as the mechanism of rice symptom development.What's more, the result is useful for designing efficient targets when attempting to produce RNAi-mediated RGSV-resistant rice plants.
