中文摘要: 设计合成近红外荧光探针应用于生物样品的荧光分析成像在生物医学研究领域发挥着重要作用。本项目在硅取代罗丹明分子骨架中引入内酯螺环,获得一种结构新颖的荧光分子,它既具有生物适用的近红外光谱特性;同时又保留了传统罗丹明结构和性能上的诸多优点,如光稳定性好,荧光量子产率高,伴随颜色和荧光"关-开"变化的开环过程等。以此分子为设计平台,将适当的识别基团有机地连接到硅基罗丹明分子上,设计合成一类近红外荧光探针分子,并利用内酰胺螺环的诱导开环机理,实现分析物的选择性近红外识别传感,通过和参比罗丹明探针的比较,深入的研究硅取代对探针分子光谱性质和识别性能的影响。并进一步实现活体细胞及组织的近红外荧光成像,初步研究分析物在活体细胞及组织中的实时浓度和空间分布,为生物样品的近红外荧光成像提供新思路和新方法。

英文摘要: Design and synthesis near-infrared fluorescent probes for biological imaging plays an important role in biological and medical research. A new near-infrared fluorophore, spirolactonized Si-Rhodamine (SiR), will be prepared while retaining the other excellent characteristics of rhodamine derivatives, such as high fluorescence quantum yield, excellent photostability and unique spirolactam ring-opening process accompanied by chromogenic and fluorogenic "turn-on" signals. SiR will act as a general and robust platform to construct a series of novel NIR fluorescent probes by coupling appropriate ligands on spirolactam ring. The Si-rhodamine-based NIR probes will be applied for sensing various analytes with high sensitivity and selectivity utilizing the analyte-induced spirolactam ring-opening process. By comparing with the traditional rhodamine-based probes, the distinction of spectral properties and binding abilities evoked by the substitution of silicon will be deeply studied. Furthermore, we expect that these probes would be applicable for monitoring analytes in living cells and organisms and provide new strategies and approaches for biological imaging.
