中文摘要: 脑缺血再灌注损伤是缺血性脑卒中发生神经损伤的关键因素,早期神经细胞保护及后期的神经修复一直是该领域的研究热点。Ghrelin是近年来首先从胃组织中发现的内分泌活性肽,其受体(GHSR Ia)在中枢和外周广泛分布,参于多种生理和病理过程。从2005年我们首先报道了其对脑缺血再灌注损伤的神经保护作用以来,受到广泛的关注,已积累了很多研究资料。然而,从神经血管单元(NVU)角度综合探究其神经保护机制,尚未被关注。Ghrelin在脑缺血再灌损伤后期神经修复中的作用也未见研究报道。本项目拟以受损脑区的NVU成员(神经元、星形胶质细胞和微血管内皮细胞)之间相互网络联络为靶标,深入探讨ghrelin的神经保护机制,特别是其在脑损伤后期的神经修复过程中的整合调控作用,阐明介导其作用的关键修复分子。研究结果将对脑缺血再灌注损伤的有效的神经保护和神经修复提供新的思路,具有重要的意义和研究价值。

英文摘要: Cerebral ischemia-reperfusion injury is a key injury factor for ischemic stroke cause nerve damage, early neuroprotection and late neuronal repair has been a hot topic of in the field. Ghrelin was first discovered in recent years from the stomach tissue of endocrine active peptide receptor (GHSR Ia) in the central and peripheral widely distributed , participate in a variety of physiological and pathological processes. We first reported the neuroprotective effects of ghrelin on cerebral ischemia reperfusion injury since 2005, a lot of research data has accumulated, However, the neuroprotective mechanism of ghrelin has not been concerned According neurovascular unit (NVU)NVU views. The role of Ghrelin on repair after cerebral ischemia-reperfusion injury in the latter part have not been reported. The project intends to study the role of ghrelin on NVU (neurons, astrocytes and microvascular endothelial cells) network during neuronal repair stage after ischemia and reperfusion injury, and further explore the mechanism of ghrelin during neuronal repair in later stage, to clarify the key repair molecules that mediated the repair role of ghrelin. The findings will provide new ideas for effective neuroprotection and repair from cerebral ischemia and reperfusion injury with an important significance and research value.
