中文摘要: 本课题以当归补血汤为模型药物,采用正交设计多指标法优选出其最佳提取工艺:取处方药材加12倍量水,提取3次,每次1.0小时。根据液体分散体系的性质,研究了当归补血汤提取液的相状态,发现该体系含以均相状态存在的真溶液和非均相状态存在的胶体溶液、混悬液或乳浊液,为混合分散体系。在此基础上,参照不同分散体系的研究方法,以水为分散介质,对黄芪甲苷、阿魏酸、黄芪皂苷、黄芪多糖等当归补血汤的代表性成分与组分及当归补血汤单味药、复方提取液的理化特征进行表征,建立了包括表面特性、电性质、流变性、热力学等性质的表征方法,发现表面张力主要受黄芪甲苷类成分影响,pH值和电导率主要受阿魏酸类成分影响,渗透压可能主要受多糖类成分的影响。以当归补血汤的益气补血功效为依据,针对环磷酰胺所致血虚小鼠模型和复合因素所致气虚小鼠模型,研究了理化特征参数与药效之间的相关性,发现表面张力、电导率和渗透压这三个参数能反映当归补血汤的药效强弱。同时,采用多项式回归分析,发现当归补血汤的"理化表征参数-组分含量-益气补血药效"三者之间呈一致性变化,即理化参数与成分、药效间存在内在的必然联系,通过理化参数能够表征当归补血汤的质量。

英文摘要: Using Dangguibuxue decoction as a model drug, the project studied preparation technique by orthogonal test and the optimal were as follows: the dried herbs were boiled three times with 12 times of water for 1 h by orthogonal design. According to the properties of liquid disperse system, the phase state of Danggui Buxue decoction was studied, which was composed of the true solution in homogeneous state, and colloidal solution, suspension or emulsion in heterogeneous state. On the basis, referring to research methods of different dispersion system, using water as dispersion medium, physicochemical characterization of Dangguibuxue decotion and its single medicines and main active ingredients, such as astragaloside IV, ferulic acid, astragaloside, polysaccharides were studied, including the surface properties, electrical properties, rheological property and thermodynamic properties.It was found that surface tension was mainly affected by astragaloside IV, pH value and conductivity were mainly affected by ferulic acid and osmotic pressure might be mainly affected by polysaccharides. In addition, under the effect of invigorating Qi and enriching blood of Dangguibuxue decoction, aiming at blood deficiency model mice induced by injecting cyclophosphamide and qi deficiency model mice induced by compound factors, relationship between physicochemical parameters and effect of Dangguibuxue decoction was studied. Surface tension, conductivity and osmotic pressure could reflect the effect of Dangguibuxue decoction. At the same time, through polynomial regression analysis, results showed that physicochemical parameters closely correlated with component content and effect of invigorating Qi and enriching blood of Danggui Buxue decoction. These results preliminarily suggested that physicochemical parameters could be used to characterize the quality of decoction.
