Based on the current research progress of neural circuit in major depressive disorder and new findings of our relevant studies, we put forward a hypothesis that depression was induced by psychological cognition bias and abnormal of cognition-related neural circuit on the basis of depressive emotion. Cognitive disfunction related neural circuit plays a vital and decisive role in the pathophysiology of depression. We plan to use multimodal neuroimaging, ERP, SPECT and cognitive psychological measurements to make a comparison among depression patients with and without cognition bias before and after treatment, first-degree relatives and healthy controls, try to 1)clarify the characteristics of neural circuit, especially brain regions of cognitive activity in depression, 2)reveal whether it is an endophenotype of depression, 3)make sure the function of cognition-related neural circuit in the pathophysiology of depression, 4)establish the quantitative description model of structure and function of neural circuit with clinical pathological markers of depression and 5)provide an important intermediary target to study the interaction of gene and environment in depression from the aspects of brain function, structure and connection.
