中文摘要: 桑树是一种生态经济型林木,其抗旱、抗涝、耐贫瘠、富含重要次生物质等优势性状分子机理急需阐明,但桑树分子生物学基础薄弱,为此家蚕基因组生物学国家重点实验室牵头实施单倍体川桑基因组测序。分离和鉴定突变体基因是研究基因功能的最可靠方法,但桑树目前缺乏突变体库。针对单倍体川桑的自身特点及当前研究现状,我们采用特异诱导转座激活标签的方法构建单倍体川桑突变体库,课题的实施将获得批量的单倍体川桑突变体,克隆突变体基因,为阐明桑树重要优势性状提供新的线索,为快速、有效推进我国桑树功能基因组研究奠定基础。

英文摘要: Mulberry is an ecological economic tree with limited molecular biological knowledges. The mechanism under its important traits in drought resistance, rich in secondary metabolites as well as the tolerances in water-logging and poor soil is still unclear. The Morus genome was sequenced last year using Morus notabilis C. K. Schneider (Chuansang), which was carried out by the researchers in the Silkworm Genome Biology State Key Laboratory. Isolation and characterization of mutant alleles is the widely used way to determine the function of a gene. However, there is no mutant library in the mulberry. In the present study, we will construct a mutant library of Chuansang using specific induction of Ac-DS transposon activation tagging system. The availability of the mutant library of Chuansang will enable cloning the mutant genes, which provide a new way for understanding molecular genetics of mulberry important traits. The results obtained from this research will facilitate the functional genomic studies of mulberry.
