中文摘要: 多药物可控程序释放是药物缓释所面临的新的挑战。层层组装技术具备灵活性、可设计性、膜组成和结构精确可控的特点,将为实现这一目标提供一条有效途径。其中的关键问题是如何获得具有高负载能力的复合膜,并能够对层间的复合程度进行调控。本项目拟采用可生物降解的聚合物复合物为构筑基元来快速制备具有高负载能力的层层组装膜,并通过两种方式来调控层间的复合程度,最终实现多种药物在膜内的精确定位和可控程序释放:一种是选用结构稳定性好、扩散能力低的交联聚合物复合物为药物缓释的载体,另一种是增加基于共价键的层层组装膜或可自交联的自组装单层膜为阻隔层。此外,在进行释放动力学调控的基础上,用细胞实验来评估药物之间的协同作用的效果。本项目的研究可为药物缓释在生物医疗和组织工程领域的应用提供帮助。

英文摘要: One of the challenges of drug delivery is controlled programmable release of multi-agents. The layer-by-layer (LbL) assembly technique, a versatile way for designing and fabricating films with precisely controlled composition and structures, is going to provide a promising way to achieve this goal. The key issues are how to fabricate films with high loading capacity and how to manipulate the mixing of layers. In this project, biodegradable polymeric complexes are taken as the building blocks for fast-fabricating of LbL films with high loading capacity. Furthermore, the mixing of layers will be tailored by two different ways in order to set the therapeutics at certain positions in the films and release them in a controlled programmable way: on the one hand, cross-linked polymeric complexes with high structural stability and low diffusion ability are taken as the vehicles for multi-agent drug delivery; on the other hand, barrier layers of self-crosslinked monolayers or LbL assembled multilayers based on covalent bonding are added. Besides, cell culture studies will be carried out in order to evaluate the synergetic effect of drugs on the basis of regulating the kinetics of release.
