中文摘要: 呼吸气体分析(Breath Analysis)研究对实现重大慢性疾病的无创检测和筛查有着重要意义。但目前呼吸气体分析研究领域的发展主要存在两大挑战:一是对确定的呼吸生物标记物,与现有的临检参数缺乏定量相关性,须深入研究以实现临床诊断;二是许多疾病的生物标记物还有待研究,需要发现用于其诊断的新标记物。这都需要一种高灵敏度的实时在线分析技术,在极短的时间内完成大量呼吸样品研究。相比质谱技术,光腔衰荡光谱(CRDS)技术具有高灵敏、可携化、实时在线等优点,有着广阔的应用前景。本项目提出的基于光腔衰荡光谱技术的呼吸生物标记物研究,将重点围绕重点围绕丙酮在紫外波段的CRDS光谱库、研究呼吸气体生物标记物丙酮与血糖的定量相关性、发现重大疾病新的可靠的呼吸生物标记物,为Breath Analysis领域的研究提供基础数据和科学依据。

英文摘要: Breath analysis is an emerging research field, which can potentially lead to noninvasive, low-cost disease/cancers diagnostics and early screening using a specific breath biomarker. Current breath analysis research embraces a commondrawback that the reported clinical studies either used a limited number of humansubjects (research data insufficiency) or took a long research period in order toobtained more data (low data collecting efficiency). Technologically, this issueis due to the constraint of the current breath analysis technology ,which are predominantly based on GC-MS or MS-based methods in general. The GC-MS or MS-methods are time-consuming and expensive. They need acomplicated sample preparation, such as sample collection and pre-concentration.Consequently,breath analysis research suffers from data insufficiency. This phenomenon hinders the further advancement of breath analysis research. We propose to develop a portable, highly-sensitive, real-time, on-line breath analyzer using cavity ringdown spectroscopy (CRDS). The advanced laser spectroscopy will help address the issues of the data insufficiency in clinical breath analysis due to its unique capabilities of real-time response, in-situ deployment,freedom from sample pre-concentration and measurement calibration. The project brings up an investigation of breath biomarker based on cavity ringdown spectroscopy. This will provide important fundamental data and sciectific evidence for the research of breath analysis.The research idea is based on the PI's previous studies in breath analysis and his expertise in CRDS and its instrumentation.
