中文摘要: 电子断层三维重构技术是近几年迅速发展起来的研究生物大分子纳米分辨率三维结构的前沿技术,是联接细胞生物学和结构分子生物学研究的重要手段。但是,目前电子断层三维重构技术存在如下缺陷:1)缺乏解决电子断层中存在的缺失楔问题的三维重构模型;2)缺乏面向高分辨率的电子断层三维重构算法;3)缺乏高效的电子断层三维重构并行算法,重构过程极其缓慢。 针对当前电子断层重构研究存在的问题,本课题希望从三个方面对面向高分辨率的电子断层重构算法开展研究:第一,研究新的电子断层三维重构模型,尽可能弥补缺失数据对重构结果的影响;第二,在新重构模型基础上,研究高精度的重构算法,提高三维结构的分辨率;第三,针对高分辨率的电子断层数据,研究其在多GPU上的存储策略和多GPU之间的数据通信机制,开发基于多GPU平台的高效电子断层三维重构算法,进一步提高重构算法的性能。

英文摘要: Three-dimensional (3D) reconstruction of electron tomography (ET) has developed rapidly in recent years as a leading technique in analyzing structures of complex macromolecules in nano scale. It has been an important method to combine cell biology with structural molecule biology. However, it is difficult to obtain high-quality reconstructed results in ET due to the following reasons: 1) There are few 3D reconstructing models to resolve the problem of the missing wedge existing in ET. 2) 3D reconstruction algorithms recently used in ET cannot obtain high-resolution reconstructed results. 3) current parallelizations of 3D reconstruction of ET are not effective and reconstruction processes are still time-consuming. To resolve the problems existing in ET, we improve the performance of 3D reconstruction algorithm in ET from three aspects as follows: 1) We will study new 3D reconstruction models to resolve the problem of the missing wedge in ET. 2) Based on the new reconstructing models, we will develop effective reconstruction algorithms to improve the quality of reconstructed results. 3) To handle large scale ET data effectively, we will study storage strategies and communication schemes on multi-GPUs, and develop an effective parallel strategy of 3D reconstruction in ET on multi-GPUs to exhibit a significant acce
