中文摘要: 相较于其祖先假结核菌,鼠疫菌演化出经跳蚤为媒介的独特传播途径,且与鼠疫菌形成生物膜紧密相关。已有研究表明:鼠疫菌的rcsA基因在进化中成为假基因,因此无法表达RcsA而不能形成复合体型调控子RcsAB而去抑制生物膜基因的转录,从而使鼠疫菌具备了在蚤体中形成生物膜的能力。本研究试图探究调控子RcsB和RcsAB对鼠疫菌生物膜的调控作用,探究调控子RcsB和RcsAB对生物膜基因hmsHFRS、hmsT、hmsD、waaA的转录抑制作用,深入研究RcsA和RcsB蛋白在基因调控中的单独与协同作用机理,了解RcsAB调控环路变化与鼠疫菌生物膜形成能力进化的关系,以进一步阐述鼠疫菌经媒介传播的分子机制。

英文摘要: Compared to its ancestor Yersinia pseudotuberculosis, Yersinia pestis (the causative agent of plague) has evolved the flea-borne route of transmission, and the biofilm formation of the plague bacteria greatly contributes to the transmission. As shown previously, the rcsA gene has become a pseudogene in Y. pestis, which makes the absence of the RcsAB complex that acts as a transcriptional repressor of biofilm genes; in turns, the plague bacteria evolves the ability to form biofilms in flea vector. In this study, the transcriptional regulation of the biofilm phenotypes and the biofilm genes (hmsHFRS, hmsT, hmsD, and waaA) by RcsB and RcsAB, and the interaction of RcsB and RcsA in gene regulation will be characterized to explore how the changes in the regulatory circuits controlled by RcsAB promote the evolution of biofilm formation in Y. pestis. The results will the promote the deeper understanding of the molecular mechanisms of the flea-borne route of transmission of Y. pestis.
