中文摘要: MicroRNA能调控细胞的增殖、凋亡、转移等多种生物学行为,在肿瘤的发生发展中具有重要作用。miR-155在许多肿瘤中均呈上调表达,可促进肿瘤发生,但gga-miR-155在马立克氏病病毒(MDV)转化的T淋巴细胞系中呈下调表达,目前还不清楚其在MDV致瘤中的具体作用。因此,本研究拟以gga-miR-155为切入点,在MDV 转化的T淋巴细胞系MSB1中实现gga-miR-155过表达和抑制表达的基础上,检测其对细胞增殖、凋亡、迁移和侵袭等生物学行为的影响,同时在mRNA水平、蛋白水平检测靶基因RORA及其相关信号通路Wnt/β-catenin关键信号分子的表达水平和活性变化,验证其与MSB1细胞生物学行为的相关性,阐明gga-miR-155通过RORA/Wnt/β-catenin通路调控MSB1生物行为的作用机理。研究结果将为马立克氏病的分子靶向预防奠定重要的理论基础。

英文摘要: MicroRNAs play an important regulatory role in various aspects of cell biology, including proliferation, apoptosis, migration and tumorigenesis. The expression of miR-155 promoting the tumorigenesis was up-regulation in neoplasm, but the expression of gga-miR-155 was down-regulation in Marek’s disease virus-transformed T-lymphocyte lines, however, the effect of gga-miR-155 on the tumorigenesis of MDV remains unclear. Therefore, the present study will examine the effect of gga-miR-155 on the biological behavior including proliferation, apoptosis, migration, invasion of MSB1 through gga-miR-155 over-expression or suppression expression by knockout or introduction of exogenous gga-miR-155. The changes of the expression and activity of RORA and the key signal molecules of Wnt/β-catenin, and the effects on the biological behavior of MSB1 will also be studied. The mechanism of gga-miR-155 on the biological behavior of MSB1 by the signal pathway of RORA/Wnt/β-catenin will be further illuminated. The findings will provide a significant theoretical basis to prevent marek’s disease.
