中文摘要: 研究表明硒元素与衰老及衰老相关疾病密切相关,近年来备受瞩目。β-乳球蛋白一度被认为是乳酪、干酪等乳制品加工的废弃物。将β-乳球蛋白与SeO2化合制备新型亚硒酸乳球蛋白,将成为一种价廉、安全、易开发的新型有机硒产品,有利于硒和β-乳球蛋白的共同开发利用。本项目主要将β-乳球蛋白与SeO2等比共溶,在真空低温条件下通过氧化-还原将亚硒酸导入到β-乳球蛋白的精氨酸残基的氨基上得到亚硒酸β-乳球蛋白。利用荧光光谱、圆二色性、红外光谱、1H和13C核磁共振光谱、电喷雾质谱(ESI-MS)、X-射线衍射等技术研究亚硒酸β-乳球蛋白的空间结构和真空条件下β-乳球蛋白的空间结构。采用免疫组化、免疫印迹、原位杂交、RT-PCR、免疫荧光、流式细胞仪、激光共聚焦等技术研究亚硒酸β-乳球蛋白的抗衰老机制。本研究不仅对有机硒化蛋白的研制具有重大理论意义,而且对"健康老龄化"具有重大的社会现实意义。

英文摘要: Selenium(Se) is an essential element for organisms. Interest in selenium-containing therapeutics has grown over in recent years. The beneficial effect of Se is not only dose-dependent, but also relates to the chemical form of the element and its bioavailability. It is strongly suggested on the basis of a higher retention and a lower toxicity, that organic Se is more recommendable than inorganic Se in supplementation. β-lactoglobulin is the major whey protein of ruminant species containing 3 arginine residues. In the study, a novel seleno-β-lactoglobulin will be synthesized at low temperature under vacuum conditions using our unique patent technology. Conformational changes of β-lactoglobulin under different reactive conditions will be investigated. The mechanism of selenium dioxide (SeO2) specific binding to arginine residues in β-lactoglobulin will be analyzed using fluorescence, circular dichroism, FITR, 1H NMR,13C NMR, ESI-MS and X-ray crystallography technologies. In recent years, selenoproteins are thought to be played important roles in preventing aging and aging-associated diseases, e.g. Alzheimer's disease (AD) and Parkinson's disease (PD).
