中文摘要: 职业暴露流行病学调查显示,二甲基甲酰胺(DMF)对肝脏具有不同程度的损伤。然而,长期低浓度DMF暴露,是否对非职业暴露人群肝脏也具有一定损伤作用及DMF致肝损伤机制至今不明。本项目拟通过对二甲基甲酰胺(DMF)重污染区非职业暴露人群肝病住院情况的全年统计调查,利用时间序列分析法,探讨DMF人群暴露浓度与肝损伤之间的相关性,确立暴露响应浓度范围。在前期研究基础上,根据响应浓度范围,利用整体动物吸入式暴露实验,研究DMF对小鼠肝脏中长期低浓度下肝脏损伤情况。进一步结合体外细胞实验研究DMF诱导肝损伤的可能机制。主要运用高通量芯片及免疫组化,蛋白印迹等生物学技术关注DMF对肝细胞凋亡诱导上的相关分子信号通路。拟揭示DMF在环境浓度下对肝脏的损伤及可能的相关机制。为DMF非职业暴露人群的影响评价填补空缺;为DMF使用、管理等提供毒理学数据支撑。

英文摘要: Epidemiology study on occupational workers clarified that dimethylformamide (DMF) was reported to have hepatic injury. Information relate on the hepatic effect on general population under long term low dosage exposure, however, is not available. The study intends to investigate the relationship between DMF exposure and hospitalization for different types of hepatic diseases in general population by time series study. Further, the mechanism for the DMF induced injury will clarify both on in vitro cellular model and animal exposure assay. The mechanism research is mainly focusing on the signaling pathway of hepatic cellular apoptosis induced by DMF through microarray, immunohistochemistry, and western blot, etc. methods. This study will supply the void of risk assessment of DMF on general population and give the toxicological support for the further usage and management on DMF.
