中文摘要: 由于在细胞成像和临床检测等领域的潜在应用,具有双光子诱导荧光的稀土配合物近年来成为人们研究的热点。项目拟进行长波敏化水溶性稀土配合物的设计、合成及双光子细胞显影研究。以2,2′:6′,2?-三联吡啶作为电子受体,利用膦叶立德Wittig反应和Sonogashira偶联反应分别构筑双键型和叁键型D-π-A共轭分子,以Eu(III)和Tb(III)为发光中心,设计合成系列水溶性高双光子活性稀土配合物。研究配体及稀土配合物的单/双光子光学性质以及稀土敏化过程中的能量传递机制;研究电子给体对配合物敏化波长的影响;通过量子化学计算揭示分子结构与荧光性能之间的关系;利用电喷雾电离质谱,研究配合物在水溶液中的稳定性;以宫颈癌细胞为对象,研究配合物的细胞穿透能力、毒性;评估配合物在双光子细胞成像领域的应用。本方案可解决现有水溶性荧光探针双光子活性系数小,敏化波长低的缺陷,具有重要的科学意义和研究价值。

英文摘要: Research into two-photon absorption induced luminescence of lanthanide complexes has become increasingly intensive because of their potential application in cell imaging and clinical examination. In this project, the design, synthesis, and two-photon absorption induced luminescence properties of water soluble long wave-sensitized lanthanide complexes for live cell imaging will be discussed. With 2, 2′: 6′, 2?-terpyridine as acceptor (D), π-conjugated D-π-A type derivatives will be designed and synthesized by Phosphonium Ylids Wittig reaction for double-bond derivatives and Sonogashira coupling reaction for triple bond derivatives, respectively. Then, lanthanide complexes with high TPA coefficients will be designed and synthesized using Eu(III) and Tb(III) ions as luminescence center. The single and two-photon absorption properties of the ligands and the corresponding lanthanide complexes, the energy transfer mechanism in the sensitization process, the influence to the wave length from the electron donors will be researched, together with the structure-property relationship by means of computational methods of quantum chemistry.
