中文摘要: 脊髓损伤是一种多发病,预后差。尚无有效的临床治疗方法。我们在前期研究的资助下,发现PPAR-γ的激活可抑制脊髓损伤后的炎症反应及促进神经干细胞的增殖,但同时也抑制了NF-κB的表达及神经干细胞向神经元的分化。最近有国外研究发现:NF-κB 及其下游的PKA,与神经元的分化及神经轴突的形成密切相关。据此我们假设PPAR-γ的激活在抑制脊髓损伤后NF-κB/PKA表达的同时,也抑制了神经元的定向分化和轴突的形成。我们拟在体外神经干细胞培养体系中通过腺病毒载体的构建及转染等,收获具有PKA高表达及低表达的神经干细胞。在体内试验中,激活PPAR-γ途径后,将体外收获的高表达及低表达干细胞分别移植入脊髓损伤动物模型中,探讨在脊髓损伤修复过程中炎症的表达,神经元、神经网络的形成以及PPAR-γ 、NF-κB/PKA通路之间的联系,为治疗脊髓损伤提供新思路。

英文摘要: Spinal cord injury (SCI) is frequently encountered disease with bad prognosis and few effective clinical therapies. With the support of research before, we found the activation of PPAR-γ could inhibit the inflammation and increase the neural stem cell proliferation, and inhibit the expression of NF-ΚB and neuronal differentiation. According to recently study aboard, NF-κB and downstream factor PKA is involved in neuronal differentiation and axonal forming.Accordingly,we assume that the activation of PPAR-γ not only inhibit the inflammation after spinal cord injury,but also restain the euronal differentiation and axonal forming by inhibiting the NF-κB activation.We are prepared to construct and transfect Adeno-virus vector in vitro neural stem cell culture system, and harvest PKA high expression and low expression of neural stem cells.With an agonist of PPAR-γ,we activate PPAR-γ pathway . Then analyze the inflammation,the neural stem cell proliferation and the forming of neuron and axon. In the same time, we will investigate the critical control factor and the connection between two signal pathway
