中文摘要: 目前对于挥发性麻醉药后处理的实验研究绝大多数是在正常动物模型上进行的,而临床上面临心肌缺血的病人常常存在着其他的病理状态,例如高胆固醇血症等。因此挥发性麻醉药后处理要引入临床,必须要在疾病动物模型上进行临床前实验。本项目组在正常动物进行了大量研究的基础上,由此提出了七氟烷后处理对于病理性动物心脏的作用问题。同时利用我们在在体和离体心脏研究方面理论知识和实验技术的积累,通过建立高胆固醇血症大鼠模型,来评价七氟烷后处理对于高胆固醇血症大鼠心肌的作用效果。新型膜修复蛋白MG53是新发现的仅存在于横纹肌细胞的重要膜修复蛋白,参与了心肌缺血再灌注损伤的负性调控,本课题同时深入探讨了MG53在七氟烷后处理中的作用及其与我们既往研究的PI3K/Akt途径和MPTP等机制间的联系。通过本项目研究,希望初步为挥发性麻醉药后处理进入临床试验或者临床应用提供有益的提示和理论依据。

英文摘要: Now,most animal researches about volatile anesthetic postconditioning used normal objects, but a lot of patients with myocardial ischemia were accompanied by other pathological conditions, such as hypercholesterolemia, etc.. Therefore, we must carry out preclinical animal experiments on sick animal models, if we plan to employ volatile anesthetic postconditioning in patients. Our research group proposes to study the effect of sevoflurane postconditioning on pathological myocardium, after we had conducted intensive researches on this fields. Here, this project investigates the effects of sevoflurane postconditioning on myocardium with hypercholesterolemia. MG53, a newly identified TRIM-containing family protein primarily expressed in cardiac and skeletal muscles, has been implicated in myocardial I/R injury regulating. Consequently, we also explore the role of MG53 in sevoflurane postconditioning and the relations among MG53, MPTP and PI3K/Akt pathway, after we had prepared the theoretical knowledge and experimental technique for several years. Our research group expects to gain beneficial enlightenment and academic groundwork before clinic trials or applications of sevoflurane postconditioning are launched.
