中文摘要: 为阐明中药汤剂治疗疾病的机理,探索汤剂药效物质基础化学成分体内外变化过程的科学规律。本课题采用汤剂中使用频率最高的甘草为研究载体,对其可检出的多种化学成分在甘草单味、及其组成的药对、复方三个层面的体内外动态变化过程进行多成分同时监测比较研究。研究目标是,第一、对可检出多成分同时、动态监测,研究单味药多成分间的相互影响,分别在不入血和可入血、原形入血和转化型入血的化学成分中明确药效物质基础;第二、在单味药研究的基础上,在药对层面上,考察药对中另一味中药对原单味药的可检出多成分的入血、转化等过程的影响,探索药对与单味药的药效物质基础化学成分的差异;第三、在单味药和药对研究的基础上选取经方,在复方层面上,进一步考察组成复方的其他中药对原单味中药的药效物质基础化学成分的影响;第四、优化实验流程,力争推广建立中药汤剂可检出多成分药效物质基础实验方法路线。

英文摘要: To clarify the mechanism of Chinese medicine and the changes of chemical composition in vitro and in vivo.We compared the dynamic process in vivo and in vitro of chemical constituents on the level of licorice,drugs compatibility and compound prescription.We established the HPLC fingerprint of rats' serum samples from hepatic portal vein, serum samples from aorta abdominalis and samples of intestinal absorption of Glycyrrhiza uralensis Fisher to monitor the changes of chemical composition.On this basis, using the established methods, analyzing and comparing HPLC fingerprint spectrum of serum samples, the sample of herb, the sample after intestinal metabolism, rats' serum samples from hepatic portal vein and rats' serum samples from aorta abdominalis in order to infer the metabolism of components.In the level of drug compatibility,we studied the changes and conversions of chemical composition in vitro and in vivo after adding other herbs.Then we studied the changes and conversions of chemical composition in vitro and in vivo in the base of compound prescription.Lastly,we wanted to optimize the experimental process,and strive to promote the establishment of the experimental method route of detecting multi-component pharmacodynamic material of chinese medicine decoction.
