中文摘要: 癌症是严重危害人类健康的疾病,低毒、高效抗癌药物是当前研究热点。本项目在课题组前期研究基础上,基于甾体内酰胺甾核具有提高甾体化合物细胞毒性和降低自身毒性、而B-nor甾核可以大大提升化合物细胞毒性的特点,设计合成二类具有特殊甾核结构的甾体杂环化合物。其中一类甾核具有A-homo内酰胺结构及B-homo内酰胺结构,17-支链为不同的芳香杂环药效团;另一类为具有B-nor-[6-5-6-5]甾核结构及胆甾支链,在甾核6-位引入不同的芳香杂环药效团。同时,采用MTT法对所合成的化合物进行抗肿瘤活性筛选、对体外效果明显的化合物进行裸鼠体内实验并进行构效关系研究,同时建立具有特殊甾核结构甾体杂环化合物库;利用流式细胞仪研究化合物诱导肿瘤细胞凋亡情况并采用荧光光谱等手段研究化合物与蛋白质相互作用机制,争取在甾体化合物作为抗肿瘤药物研究方面取得新突破,为开发出具有自主知识产权的新型抗癌药物提供理论基础。

英文摘要: Cancer is a serious disease damaging human beings health. Research of effective anticancer drugs with low toxicity is a popular topic in present. Base on the distinct cytotoxicity and low toxicity of the steroid lactams and B-nor steroids found in our preliminary study, series of steroidal A-homo and B-homo-lactams possessing the heteroaromatic compounds in side chain and B-norcholesteric derivatives with the heteroaromatic in 6-position of steroidal nuleus were designed and synthesized in this proposal. The anti-tumor activity of the synthetic compounds were evaluated by MTT method and the test in animal model were assayed for the active compounds. Apoptosis of the compounds were determined using a flow cytometry. The interaction mechanism of compounds and protein were studied by fluorescence spectrum. The results generated in the studies is valuable for the design of novel chemotherapeutic drugs.
