中文摘要: 癌症治疗是人类面临的重大健康难题之一,发展聚合物类抗癌药物纳米载体是攻克这一难题的重要方向。本项目拟采用功能性的多肽链作为反应单体合成多肽类聚合物,使其本身同时具有高效的抗癌效果及良好的生物相容性。以不同功能性的多肽为研究对象,如靶向肽、穿膜肽、定位肽及治疗肽等,采用迈克尔加成的方法聚合得到具有多种生物功能的多肽共聚物;并研究刺激响应性连接基团的引入对多肽分子在肿瘤部位的可控释放行为的影响。通过高通量的合成及筛选,揭示多肽聚合物的组成和结构与其生物学性质的关系,最终发展得到具有高效癌症治疗效果的多肽共聚物。通过本项目的实施,拟发展功能性多肽聚合物合成的新方法,建立其化学结构与生物学性质的关系,为发展新型大分子纳米药物提供新思路和新途径。

英文摘要: Cancer therapy is one of the important human health problems.The polymer therapeutics conjugated with anti-cancer drugs have been an important drug nanocarriers. The macromolecular therapeutics can be retained in tumor sites due to the enhanced permeability and retention effect (EPR effect). However, these synthetic polymer drug carriers have potential bio-toxicity and immunological rejection, so we proposed a group of peptide polymers which have enhanced cancer therapy effect and good biocompatibility. Peptides have many biology functions such as targeting, therapy, cell-penetrating, localization and so on. The multifunctional peptide polymers can be prepared by copolymerizing various functional peptides. The copolymers with different compositions and structures can be obtained by varying the type of peptides and linkers. In addition, introducing stimuli-sensitive linkers can realize the controlled release of peptides at tumor sites. Various peptide copolymers can be high-throughput synthesis and screening to obtain copolymers with enhanced cancer therapy effects by cell biology measurenments.Therefore, the project will develop the new synthesis method of peptide polymers, study the relationship between chemical structure and biology propeties, and provide new approach for developing novel macromolecular nano-d
