中文摘要: 抑制蛋白酪氨酸磷酸酶1B(PTP1B)的活性是有效治疗糖尿病的一条途径。虽然报道过不少的合成PTP1B抑制剂,但由于它们自身的原因而不适于成为候选药物。因此,越来越多的科研人员着眼于从自然界获取PTP1B抑制剂。海洋中的圆盘肉芝软珊瑚能产生很多结构新颖、生物活性显著的次级代谢产物,是当前药物研究开发的一个重要资源。最近,我们在从海洋生物资源中寻找新型药物先导结构的过程中,对采自海南的圆盘肉芝软珊瑚进行了初步的研究,从中得到了多种骨架的结构新颖的二萜化合物,并经生物活性筛选实验证实具有显著的PTP1B抑制活性,有望成为新的天然药物先导结构。为此,本项目拟在前期研究基础上,尝试对采自不同海域的圆盘肉芝软珊瑚进行系统、深入的化学成分及生物活性研究,鉴定并表征新颖二萜的结构,筛选出高效、低毒的PTP1B抑制剂先导物,研究构效关系。

英文摘要: Inhibition of protein tyrosine phosphatase 1B (PTP1B) activity is an effective way to prevent diabetes. Although there are many synthesized PTP1B inhibitors, they can’t be drug candidates due to their own properties. Therefore, more and more researchers pay their attentions to natural PTP1B inhibitors. Marine soft coral Sarcophyton trocheliophorum can produce a lot of secondary metabolites with novel structures and biological activities, which is an important resource of drug leads or medicinal candidates. Recently, in our efforts to search biologically active substances from marine sources, a series of novel diterpenes with different skeletons were obtained from the Hainan soft coral S. trocheliophorum by applicant. In the bioactive assay, these compounds showed significant PTP1B inhibitory activity. For the inspiring pioneer investigations, the in-depth chemical study of S. trocheliophorum collected from different areas, structural characterization of novel diterpenes and preliminary analysis of structure-activity relationship were proposed by applicant in order to find new bioactive natural PTP1B inhibitory drug leads.
