中文摘要: 全氟辛酸(Perfluorooctanoic acid,PFOA)是最常见的全氟化合物(Perfluorinated chemicals,PFCs)之一,广泛用于各种工业和商业产品。PFOA的持久存在和全球性的分布使其成为毒物学研究热点。研究发现,PFOA有潜在的神经毒性、基因毒性、分子毒性、生殖及发育毒性。然而,关于PFOA对水生动物群的毒性尤其是对淡水无脊椎动物的毒性和毒性机制知之甚少。涡虫以其简单的形态和基础进化地位、极强的再生能力和对毒素的高敏感性而成为研究再生、发育和水质监测的模式生物。本项目拟以东亚三角涡虫作为研究对象,用再生发育参数、抗氧化酶活性、细胞凋亡、基因的表达、蛋白的表达等指标来评价PFOA对涡虫的影响,研究PFOA诱导的氧化应激损伤及细胞凋亡,探索PFOA对涡虫的毒性影响和可能的机制,为进一步深入研究PFOA在水生环境中的生态毒性奠定基础。

英文摘要: Perfluorooctanoic acid (PFOA) is one of the most common perfluorinated compounds (PFCs) and has been widely used in a variety of industrial and commercial products. PFOA has become a hot subject of toxicological studies because of its persistence in the environment and global distribution. Recent toxicological studies showed that PFOA has potential neurotoxicity, genotoxicity, molecular toxicity, and reproductive and developmental toxicities in animals. However, little is known on the toxicity of PFOA to aquatic animals, especially to freshwater invertebrates. Freshwater planarians are used as a suitable organism to study regeneration, development and water quality monitoring because they have simple morphology, basal evolutionary position, high regeneration capacity and high sensitivity to toxicological agents. In this study we will evaluate the toxicological effects of PFOA on the planarian Dugesia japonica using regenerating developmental parameters, antioxidant enzyme activity, apoptosis, gene expression and protein expression biomarkers. The main purposes of this study are as follows: (1) examine cellular toxicology of PFOA on oxidative stress; (2) investigate whether apoptosis may be induced; (3) explore the toxicity and mechanism of PFOA in planarians Dugesia japonica; and (4) provide the information for
