中文摘要: 近年来,癌症发病率大幅度提升;而环境中致癌物质如黄曲霉素的累积被认为是诱发的癌变主要因素。在癌变过程中,活性氧与自由基发挥着重要的作用;深入了解细胞氧化还原平衡在癌变过程中的演化对于揭示细胞癌变的机理至关紧要。目前对于黄曲霉素致癌机理的研究主要通过分析群体细胞或整个细胞的遗传学和表观遗传学变化,而亚细胞水平如不同细胞器内的氧化还原变化以及氧化状态引发的细胞信号转导仍不甚清楚。本课题拟利用黄曲霉素诱导正常肝细胞癌变,并采用一个独特的高时空分辨率的纳米光电传感器来获取癌变启动阶段的细胞在亚细胞水平上的细胞信号,以此探明细胞器内氧化剂与抗氧化剂的相互作用和整体氧化还原平衡的时空变化,确定氧化还原控制的亚细胞关键位点以及受氧化状态控制的靶分子的变化,由此揭示黄曲霉素诱发细胞癌变的机率与亚细胞氧化还原平衡的联系。

英文摘要: Reactive oxygen species(ROS) plays a critical role in the process of carcinogenesis. ROS generation has also been implicated in Aflatoxin-induced carcinogenesis. Therefore, in-depth understanding of the evolution of cellular redox homeostasis during aflatoxin induced-carcinogensis is crucial to unravel the underlying mechanism of cancer development.Currently, the research on the cellular redox changes still relies on the analysis of a population of cells or whole cells to identify the genetic or epigenetic changes. Thus, the carcinogensis associated redox changes on the subcellular level remains unclear. This project plans to apply aflatoxin B1 to induce normal hepatic cells to transform into cancer cells. An unique nanoscale electro-optical sensor will be used to detect the subcellular level cellular signals. The results obtained will reveal the spatio-temporal interplay between the ROS and the antioxidant as well as the overall redox homeostasis after aflatoxin exposure, determine the critical subcellular sites in redox control ,and also uncover the redox controlled targets such as transcription factors, finally leading to the understanding of the relationship between subcellular redox changes and aflatoxin induced-carcinogenesis.
