中文摘要: 蛋白激酶活性的实时高敏分析方法对癌症成因/进程及抗癌药物开发研究具有重要意义。本项目拟将恶性肿瘤相关蛋白激酶作为主要研究对象,设计、开发和表征具有光学、电化学或电学性质的多功能多肽底物,借助纳米/聚合物/聚电解质材料的优异性质,结合磷酸化状态诱导多肽的设计技巧,研究多肽底物磷酸化过程中自身性质变化及与多种纳米材料/聚合物/聚电解质间相互作用改变导致的光学、电化学或电学信号实时变化规律,籍此开发蛋白激酶活性的实时高敏生物传感方法,实现活细胞内蛋白激酶活性的评估及癌变细胞的异常信号捕获,并用于筛选蛋白激酶抑制剂,为癌症诊疗和抗癌药物的开发提供新方法和新知识。包括:(1)设计与合成具有信号输出单元的新型底物多肽;(2)设计新型多肽功能生物传感器用于研究蛋白激酶活性;(3)评估一些药物小分子对蛋白激酶活性的抑制能力,筛选蛋白激酶抑制剂;(4)基于新法开展癌细胞体系的蛋白激酶活性检测以及抑制剂研究。

英文摘要: The real time and highly sensitive analysis strategies towards the protein kinase activity (PKA) is of high importance for the researches of the cancer formation/development and anti-cancer drug development. This project is supposed to aim at the tumor-relative protein kinases as the main targets, design, develop, and charaterize multi-functional polypeptide substrates with optical, electronic, and electrochemical properties, introduce novel nanomaterials/conjugated polymers/polyelectrolytes with unique chemical/physical properties, combine the design skills of phosphorylation-induced peptides, explore the real time viaration regularity of the properties of multi-functional polypeptides and interactions between polypeptides and nanomaterials/conjugated polymers/polyelectrolytes during the phosphorylation process, accordingly elaborate novel real time, highly sensitive biosensing protocols of the PKA, realize the estimation of the PKA in live cell and the capture of the abnormal signal readout in tumor cell, apply to the screening of the protein kinase inhibitors, finally devote new and useful informations to the cancer diagnose/therapy research and the anto-cancer drugs developments.
