中文摘要: 基于极端环境微生物独特的生存机制和广阔应用开发前景,本项目依托高通量核酸测序技术及高通量质谱技术平台,根据基因组学、转录组学、蛋白质组学和代谢组学的研究手段,全基因组测序分析自我国新疆强光照地区分离获得的耐辐射细菌新种Pontibacter korlensis(库尔勒海洋杆菌)全基因组与已知的极端耐辐射微生物Deinococcus radiodurans(耐辐射奇球菌)进行比较分析,确定与抗辐射相关的基因。同时,从基因-蛋白质-代谢物多层次全方位研究耐辐射细菌P.korlensis和D. radiodurans在不同强度的离子辐射条件下RNA信号的变化、蛋白质表达及其相继的代谢物与代谢物表达水平的分子网络变化;发掘在以上多个层面上真正有效的抗辐射成分,揭示极端微生物迅速适应高辐射环境的分子网络调控机制和规律,系统闸明耐辐射细菌的耐辐射机理。

英文摘要: Extremophiles have great application potential due to their capacity living in extreme environment. Based on high throughput nucleic acid sequencing technology and mass spectrometer platform, the genome, transcriptome, proteome and metabolome of Pontibacter korlensis and Deinococcus radiodurans R1 will be systematicly studied. The genome of P. korlensis, a new radiation-resistant bacterium species separated from Xinjiang desert, will be sequenced and further comparatively analyzed with that of typical radation-resistant species D. radiodurans. Genes involved in radiation resistance will be identified; Furthermore, by integrating genomics, trancriptomics, proteomics and metabolomics technology, the regulatory network will be constructed through studying genes expression profiles as well as consequential proteins and metabolites changes at series of 60Co radiation intensities. This regulatory network will undoubtedly promote our later work in developing an insight into the radiation-resistant mechanism of these extremphiles and exploiting functional genes, proteins and metabolites.
