中文摘要: 原发性胆汁性肝硬化(PBC)是自身免疫与遗传易感性双重作用结果的一种自身免疫性疾病。本项目对前期480例PBC患者和480例健康对照研究发现,位于PVT1区域的SNP rs2114358与PBC有显著相关性,而该SNP刚好位于miR-1206的前体序列折叠的颈环结构中,这在目前尚属首次发现。本研究从一个与PBC易感性相关的影响miRNA分子成熟效率的SNP为研究对象,然后从分子生物学及细胞生物学水平研究该SNP对miR-1206的成熟效率影响、miR-1206靶基因的筛选,来阐述该miRNA分子对免疫细胞的影响,该研究结果将明确miR-1206在疾病进展中的作用,并详细阐明其作用机理。有助于我们进一步深入了解miRNA的多重复杂的作用机理,对一个新的角度研究PBC的发生机制、寻找新的治疗方法、改善PBC治疗效果都具有重要的意义。

英文摘要: Primary biliary cirrhosis (PBC) is an autoimmune disease which caused by autoimmunity and genetic susceptibility. We studied 480 patients and 480 healthy controls, and found SNP rs2114358 that located PVT1 area had a significant associated with PBC, While the SNP just is located miR-1206 precursor sequence folded neck ring structure. In the present, this is the first findings. In this study, we study the SNP which is associated with PBC and can affect the mature efficiency of a miRNA molecule. To illustrate the miRNA how to affect immune cells by biology and cytobiology. The results will pinpoint the role of miR-1206 with PBC progression, and will clarify the mechanism in details. Ultimately, the research will help us to further insight into the multiple mechanism of miRNA. It will be of important significance to a new perspective pathogenesis, new methods and improvement treatment of PBC.
