中文摘要: EB病毒潜伏感染、上皮-间质转化、肿瘤干细胞形成是鼻咽癌发生发展中的重要环节。鼻咽癌细胞内ZEB蛋白和miR-200形成的负反馈环,象一台持续转动分子引擎, 通过复杂的信号调控网EB病毒感染、上皮-间质转化、肿瘤干细胞形成是鼻咽癌发生发展中的三个络,推动三个环节相互促进,形成一个周而复始的恶性循环。本研究采用淋巴细胞与鼻咽癌细胞共培养技术,模拟鼻咽癌体内微环境,建立上皮-间质/溶解-潜伏动态转化细胞模型,分选具有干细胞潜能的亚群,观察ZEB/miR-200轴及其上下游信号调控网络的表达变化,分析EB病毒感染、鼻咽癌细胞上皮-间质转化和干细胞形成及干性维持影响。在此基础上,以EB病毒EBNA1基因识别的靶向载体,在鼻咽癌细胞内特异性表达miR-200,研究能否通过恢复ZEB/miR-200负反馈平衡 ,实现对EBV潜伏感染/EMT/肿瘤干细胞形成三个环节的同步干预,达到控制鼻咽癌的目的。

英文摘要: Epstein-Barr virus (EBV) latent infection,Epithelial-mesenchymal transition (EMT) and formation of cancer stem cells(CSC)are crucial factor in the pathogenesis of Nasopharyngeal carcinoma (NPC). Here we hypothesis that is closely connected to each other by ZEB/miR-200 feedback loop, a vicious circle of the three procedures is involved in sustained undifferentiation, early metastasis and resistance to chemo-radio therapy of NCP. The aim of this study is to explore the modulation of ZEB/miR-200 loop in EBV latency/EMT/CSC circle in pathogenesis of NPC. By sorting cells by CD133 and ABCG-2 with FACS, we will observe the different expression of EBV-encoded genes, EMT markers as well as cell stemness signaling-pathway, and find out if we can modulate the ZEB/miR-200 molecular switch to break the EBV Latency/EMT/CSC vicious circle. With an EBNA1 targeted lentivious vector, we can over-expressing miR-200 in EBV latent-infected NPC cells both in vivo and in vitro, and finally develop a novel therapeutic strategy of NPC.
