中文摘要: 随着纳米技术迅速发展的表面增强拉曼效应能够快速、无损、灵敏、廉价地实现生物组织、细胞的检测,包括癌组织的早期检测,在医学影像及诊断中具有巨大的应用潜力。本项目着眼纳米多层结构拉曼基底中的基础科学问题,制备生物相容的高度选择性的表面增强拉曼基底,实现小动物活体癌组织的拉曼检测,进而实现其癌组织的拉曼成像,推动表面增强拉曼效应在医学诊断中的应用。 在项目研究中采用模拟分析指导物理气相沉积特定纳米结构的高灵敏度表面增强拉曼基底,如银基底;再通过化学方法吸附拉曼标签分子,并制备包覆层以提高生物相容性;通过代谢过程的研究来关联选择性输运的分子,通过免疫学方法关联选择性吸附的分子,使得制备的表面增强拉曼探头能够提供指定生物组织的信息;最后将所制备的探头药剂注入小动物静脉,实现指定癌组织的拉曼检测并得到拉曼影像。本项目的研究也将为表面增强拉曼效应在生物组织选择性检测以及医疗影像中的应用提供研究支撑。

英文摘要: Surface-enhanced Raman scattering(SERS), which developed with nano technology is a powerful means for quick, noninvasive, sensitivity, cheap tissue and molecular detection, including cancer tissue detection. Thus, SESR has enormous potential in medical diagnosis and biomedical imaging. This project focuses on basic science issues in nano multilayer SERS substrates, fabricating biocompatible and highly selective SERS substrates, and in vivo tumor targeting and spectroscopic detection by injecting these substrates. In this project, at first, we will use physical vapor deposition directed by theoretical simulation to prepare highly sensitive SERS substrates, such as Ag substrates; then, make the substrates absorbed by Raman label molecular and coated with shell layer to improve biocompatibility; third, conjugate the coated SERS label with selective molecular, including transport selective molecular which is chosen by metabolic process analyze, and absorption selective molecular which is chosen by immunology analyze, and these conjugated SERS probes will be able to detect targeting tissue or molecular; at last, inject these SERS probes into small-animal via tail-vein, detect targeting tumor tissue, and achieve Raman imaging.
