中文摘要: IL-17F是新近发现的属于IL-17家族中的一个前炎症因子。在炎症反应、肿瘤免疫应答等病理过程中发挥重要作用。申请者新近研究发现,在实验性角膜新生血管模型中,IL-17F重组蛋白能够抑制骨髓造血干/祖细胞向病变角膜组织的迁移,从而推测其可能参与实验性新生血管性眼病发病的病理过程。因此本研究旨在通过:(1)利用IL-17F基因敲除小鼠,构建角膜和脉络膜新生血管模型。检测其在角膜、脉络膜新生血管的发生发展、相关细胞因子及其受体的表达以及炎症细胞、干/祖细胞迁移等过程中的作用;(2)利用IL-17F分子重组蛋白和中和性抗体体外刺激角膜和脉络膜血管内皮细胞,检测其在血管网形成、细胞迁移、增殖和凋亡等过程中对血管内皮细胞的作用等相关实验,综合分析和明确IL-17F在实验性新生血管性眼病发病过程中的确切作用和具体机制,为临床基于通过干预IL-17F等前炎症因子的途径治疗新生血管性眼病提供理论依据。

英文摘要: Cornea and choroid are immunoprivileged site. When it meets with infection,chemical injury,AMD or other inflammatory diseases,Corneal Neovascularization (CrNV) or Choroidal Neovascularization (ChNV) will occur and frequently leads to impaired vision. The physiologic regulation of angiogenesis dependents on the proper temporal and spatial delicate balance of activators and inhibitors present within the vascular microenvironment.In most of the preceding caused CrNV and ChNV,bone marrow derived cells,neutrophils,and macrophages infiltrate the cornea and choroid.They exert complicated roles by utilizing different chemokine receptors and proinflammatory signals in the development of CrNV and ChNV. The IL-17 cytokine family consists of 6 cytokines (IL-17A to IL-17F) and at least 5 receptors (IL-17RA to IL-17RE).And we recently found that IL-17F can inhibit HSCs migration to burned corneas and it may be involved in the process of corneal neovascularization. But the role of IL-17F involved in ocular neovascularization is still unclear.So the aims of this study are:firstly,To construct alkali-induced corneal neovascularization model and construct laser-induced choroidal neovascularization model using IL-17F relative knock out mice and explorer the effects on ocular neovacularization under the condition of lacking genes
