中文摘要: 钙离子(Ca2+)与钙通道和在睡眠-觉醒行为调控中起重要作用。在前期研究工作中,我们发现中缝背核(DRN)中的Ca2+参与戊巴比妥钠睡眠及生理睡眠的调节,在DRN,Ca2+功能的上调会抑制非快眼动(NREM)睡眠和快眼动(REM)睡眠,而Ca2+功能的下调有利于NREM睡眠。但是,目前尚不清楚DRN的Ca2+与钙通道在睡眠-觉醒行为中的调控作用及其机制。本项目将采用脑电频谱睡眠时相分析技术、免疫组化实验技术、高效液相电化学检测技术、蛋白免疫印迹、实时定量荧光PCR等多种实验方法,从整体动物的行为学、组织学、神经化学、信号传导通路,深入探讨DRN的Ca2+及L型、T-型、N-型钙通道在睡眠-觉醒行为调控中的分子生物学机制。本研究将进一步深入了解睡眠-觉醒行为的调控机制,并为临床治疗睡眠障碍疾病提供新的思路和研究方法。

英文摘要: Calcium and calcium channels play a crucial role in the regulation of sleep-wake behavior. Our previous studies indicated that calcium in dorsal raphe nucleus(DRN) takes part in both the pentobarbital induced sleep and nature sleep. The upscaling of calcium function in DRN could inhibit NREM sleep and REM sleep; while downscaling of calcium function could facilitate NREM sleep. However, it is still unknown that the effect and mechanism of calcium and calcium channels regulating sleep-wake behavior. From the view of the integral ethology, histology, neurochemistry and signal transduction path-way, this project uses EEG analysis combined with IHC, HPLC-ECL, Western-Blot and RT-PCR to investigate the mechanism of calcium and L-, T-, N-type calcium channels regulating sleep-wake in DRN. This research focusing on the regulation mechanism of sleep-wake behavior from an original perspective, will provide the theoretical bases and research methods for the development of more effective and safer drug to treat sleep disorder.
