中文摘要: 低强度光照可(LILI)对多种细胞产生复杂的生物学效应,但光生物调节作用的机制仍不明了。我们的前期研究发现发光二极管来源的低强度红光能影响间充质干细胞(MSCs)的增殖行为,其调节作用受MSCs细胞微环境的影响:即常规培养状态下增强细胞增殖能力,而添加成骨诱导因子时抑制细胞的增殖能力。该现象提示:不同微环境下LILI通过不同分子机制之间的转换而动态调节MSCs的生物学行为。因为近日节律基因同时具备光敏性和动态调节MSCs的能力,同时我们发现LILI可改变近日节律基因表达。因此本课题将观察LILI照射如何通过近日节律基因影响MSCs的细胞周期各阶段,进一步分析近日节律基因及TPKR下游信号通路如何协同影响MSCs的细胞增殖行为,并探讨节律基因在MSCs光诱导增殖过程中的作用和机制。本课题的开展将为光生物学效应的深入研究开创新的思路。

英文摘要: Low-intensity light irradiation (LILI) can influence the various biological function of a variety of cells, however the mechanism of photobiomodulation is not clear yet. Our previous work showed red light from a light emitting diode was effective in promoting various behaviors of mesenchymal stem cells (MSCs), which functionally dependent on cell microenvironments. We observed LILI only increased MSCs cellular proliferation on regular medium while it switched to decrease cellular proliferation after treated with osteogenic factors. Our finding indicates LILI dynamically regulates different biological functions of MSCs by switching within diverse mechanisms in a microenvironment-dependent manner. Circadian rhythm genes have the abilities of light sensitivity and two-way regulation of cellular proliferation. Moreover, our previous work also proved that LILI changed the expression of some clock genes This study aimed to investigate the effect of LILI on cell cycle regulation of MSCs and how it in turn promotes or decreases MSCs cellular proliferation or in specifical context. We propose TPKR signaling transduction and circadian rhythm genes play important role in biological function regulation of BMCs by LILI and the accomplishment of this proposal will shed new light on clinical and mechanism research of photobio
