中文摘要: 根据Saghai-Maroof et al. (1984) 中发表的方法,从生长于新疆的小麦类植物中提取基因组DNA,根据Appels et al. (1992)的引物序列设计小麦种植物5S rDNA 的扩增引物,进行PCR扩增,并对所得到的克隆进行序列分析。通过DNA序列进行系统发育分析,推测的所属的种间同源关系,建立一套序列集合,比较每条核酸序列所处的位置例如位置同源性比对,根据比对结果推导新疆小麦类系统发育关系。本项目研究达到的预期目标在于研究新疆小麦类缘关系较近不同种中5SrDNA的多样性,并进行分析、分类,划分到单元级;基于5SrDNA基因单元比较它们间的相似性及存在的差异;根据5SrDNA单元级的差异判断新疆小麦类之间的亲缘关系是否比其他属的亲缘关系更为接近;进一步研究这个种与其它种间可能存在的亲缘关系;对相关研究进行讨论。

英文摘要: Genomic DNA will be extracted according to Saghai-Maroof et al. (1984) from the accessions within Xinjiang Wheat species. Primers will be designed according to Appels et al. (1992). The 5s genes will be amplified by the polymerase chain reaction, cloned and sequenced and analyzed. Orthology will be speculated by Phylogenetic analysis which will be carried by analyzing the variation between 5s rDNA repeat sequences in Xinjiang wheat variety. In practice, however, in order to conduct a phylogenetic analysis with DNA sequence data, one assembles sets of sequences of putative orthology to compare individual nucleotide positions, and infer from these alignments phylogenetic relationships.Our objectives in the present study will be to (i) examine the sequence diversity of the 5S rDNA gene among the Xinjiang Wheat, (ii) classify them into unit classes, and (iii) define similarities and differences between the Xinjiang Wheat species on the basis of the 5S DNA units; (iv) assess whether Xinjiang Wheat species are more closely related to each other than to the other species of Wheat studied to date based on the different 5S DNA unit classes; (v) extend these findings to the study of possible relationships between Xinjiang Wheat species and others;
