中文摘要: 我们以前的工作发现,小鼠体内移植物的铜绿假单胞菌(PA)感染过程中,生物膜(Bf)形成及感染的程度受群体感应传导通路的调节。本项目在此基础上,运用分子克隆技术构建含绿色荧光蛋白的高表达或低表达环二鸟苷酸(c-di-GMP)的突变菌株和临床菌株,建立静止模型、持续流体模型和糖尿病鼠模型,使用组织化学、免疫荧光组化、荧光原位杂交,共聚焦激光显微镜等技术,(1)探讨c-di-GMP传导通路在未治疗的糖尿病小鼠创面形成PA-Bf过程中的调控作用,以及其对感染创面的组织形态、活菌数量、抗生素耐受性、IL-Iα和TNF-α的表达的影响;(2)探讨胰岛素干预PA运动能力、抗生素耐受性的过程中,c-di-GMP的调控作用;(3)探讨胰岛素治疗对创面形态、活菌数量、IL-Iα等指标的影响;(4)进一步在c-di-GMP通路层面上明确糖尿鼠模型病中胰岛素对PA-Bf的影响。

英文摘要: Our previous works have demonstrated that during intraperitoneal implant Pseudomonas aeruginosa(PA) infection,the biofilm(Bf) formation and the infection severity were regulated by the quorum sensing system. Based on our previous works, we will construct different strains ( with high/low second messenger c-di-GMP levels ) with green fluorescent protein by molecular cloning methods, establish various models (static、 flowing and daibetic mice models), and apply different techniques (histchemistry、immunflurescence histochemistry、fluorescence in situ hybridisation and confocal laserscanning microscopy) to these models, to 1) explore the role and mechanism of c-di-GMP signal pathway in PA-Bf formation、 histological analysis、viable bacterial count、 the expression of IL-Iαand TNF-αof the infected wound surface of untreated diabetic mice; 2) elucidate the role and mechanism of c-di-GMP signal pathway during the insulin treatment in PA motility assays、Bf formation and antibiotic tolerance; 3) investigate the effects of insulin treatment on the histological analysis、viable bacterial count、 the expression of IL-1αand TNF-α of the infected wound surface; 4) further evaluate the influences of insulin treatment on PA-Bf in daibetic mice through the c-di-GMP signal pathway.
