中文摘要: 肌酸激酶 (Creatine Kinase, CK) 是细胞能量代谢的关键酶之一,在生物体的能量代谢中起着重要作用。近年来一些研究表明,CK与其他蛋白质相互作用具有重要的生理意义。我们通过酵母双杂交实验发现CK很可能与CC2D1A有相互作用,此外生物信息学的研究表明NF-κB很可能与CK存在相互作用。由于有文献报道CC2D1A可以激活NF-κB信号通路,因此我们推测CK很可能与CC2D1A和NF-κB形成相互作用网络,CK可能在调控两者功能方面发挥重要作用。本项目拟研究CK与CC2D1A、CK与NF-κB相互作用;探索CK与CC2D1A和NF-κB的相互作用网络以及CK在这一网络中的功能。研究结果将在阐述CK与CC2D1A、NF-κB相互作用机理以外,首次揭示CK作为一个经典的能量代谢相关酶在NF-κB这一重要信号通路中的作用,有助于我们更加全面的了解CK的生理病理功能。

英文摘要: Creatine kinase (CK) is one of the key enzymes in cell energy metabolism. Recent studies indicate that the interactions between CK and other proteins have important physiological functions. In a yeast two-hybrid experiment, we found that CK interacts with CC2D1A. CC2D1A is reported to be an activator of NF-κB pathway, and bioinformatics studies indicated that CK also interacts with NF-κB. Our hypothesis is that CK forms an network with CC2D1A and NF-κB, and CK may play an important role in regulating the functions of CC2D1A and NF-κB in this network. In this project, we plan to study the interactions of CK, CC2D1A and NF-κB, and to explore the functions of CK in this network. Our studies will not only clarify the mechanism of interactions between these proteins, but also for the first time, uncover the roles of CK, a key enzyme in energy metabolism, in an important cell signaling pathway of NF-κB. It will shed more light on the physiological and pathological functions of CK.
