中文摘要: 细胞内具有特殊生理功能的活性小分子介导了恶性肿瘤和心脑血管疾病的发展过程。因此,对这些活性小分子进行实时、动态的检测将能够实现对某些疾病的早期诊断。针对当前应用于此研究的有机染料荧光探针和零维稀土配合物在稳定性方面存在的不足,本项目提出利用新型多功能材料—金属有机框架(简称“MOFs”)用于对细胞内活性小分子的检测研究。基于MOFs在结构和功能多样性调控所展示出来的优势及稀土离子自身独特的发光特点,本项目拟选用带有特异性识别基团的有机配体构建近红外发光稀土MOFs。通过MOFs中特殊基团和活性位点与目标检测物的特异性反应开发经济、灵敏、选择性的检测细胞内活性小分子的探针,并将其与细胞成像分析技术相结合,进而为相关疾病的诊断提供新手段。

英文摘要: Active small molecules with special physiological function in cell mediate the process of some malignant tumor and cardiovascular disease. Therefore, real-time and dynamic detection of them will be expected to achieve for the early diagnosis of some diseases. In view of the lack of stability for organic fluorescent probes and lanthanide complexes in this field, multi-functional Metal-organic Frameworks (MOFs) are recommended to detect active small molecules in this project. Based on MOFs in structural and multi-functional regulation advantages, and the characteristic luminescence of lanthanide ions, this project will construct near-infrared luminescent lanthanide MOFs by organic ligands with specific identification groups and active sites. Economic, sensitive and selective detection of intracellular active small molecule probes will be realized by the specific reaction between MOFs and the targets, which are expected to combine with cell imaging techonlogy and provide new tools for the diagnosis of related diseases.
