中文摘要: 花生高含油量种质资源创造和高油新品种培育是花生育种的主要方向之一。解析花生含油量形成的分子基础,对于通过分子设计培育高含油量花生品种具有重要意义。本课题以花生油脂形成的基因网络为主要研究目标,综合应用转录组学、蛋白组学、生物信息学和生物技术等多学科手段,选用本课题组培育的高油花生新品系花U606,分析其在不同发育时期油脂积累的规律和特点;综合蛋白质组与转录组比较关联分析和差异表达谱全方位分析,挖掘受转录后调控的关键蛋白/基因,研究花生籽粒发育过程中油脂形成、脂肪酸合成代谢的基因调控,解析花生油脂代谢途径与糖和蛋白质代谢途径的复杂网络及交互作用;克隆鉴定油脂形成中的关键基因,通过基因失活和超量表达等技术,分析基因网络上关键基因的功能。本研究的开展,不仅将更深刻地揭示花生油脂形成的代谢调控和分子生物学机制,还将为高油花生育种和脂肪酸改良提供宝贵的信息和基因资源。

英文摘要: High-oil peanut germplasm creation and High-oil peanut variety is one of the main direction of the peanut breeding. Parsing the molecular basis of the formation of peanut oil is of great significance for designing molecular breeding. Gene networks created in peanut oil are the main research objective. High-oil peanut U606 is selected for analyzing its regularity and characteristics of oil accumulation in different seed development, integrated application of the transcriptome, proteomics, bioinformatics and biotechnology, such as a multidisciplinary approach. Mining the key protein/gene transcription regulation, fatty acid metabolism and the pathways of complex networks and interactions; Cloning the key oil-genes. Analysis the function of key genes on gene network. This study will not only more profoundly reveal the metabolic regulation and molecular biology mechanism of peanut-oil forming process, will also provide high oil and fatty acid modified peanut breeding with valuable information and genetic resources.
