中文摘要: 内皮祖细胞(EPCs)移植治疗下肢缺血性疾病有较好的临床应用前景,但我们的临床实践和前期研究提示,糖尿病对内皮祖细胞移植促进血管新生有抑制作用。从内皮祖细胞动员、迁移、分化到新生血管形成,涉及到一系列基因的表达和细胞因子的介导,而糖尿病如何干预这一过程,其干预机制及作用环节是什么,目前不得而知。本研究旨在通过体外实验(晚期糖基化终产物及其受体(AGE-RAGE)以及相关通路阻断剂等共培养检测EPCs体外迁移、三维血管形成能力测定等)、体内实验(糖尿病鼠缺血肢体模型基础上进行各组EPCs移植,测定体内新生血管形成能力等)证实AGE-RAGE通路与EPCs血管新生功能的关系,并深入挖掘内在的分子病理机制,试图寻找糖尿病干预EPCs移植的机制与环节,为临床上提高对EPCs移植治疗下肢缺血性疾病的机制的认识,尤其是为改善其对缺血性糖尿病足的治疗效果,提供理论基础和干预契机。

英文摘要: Endothelial progenitor cells (EPCs)transplantation is a treatment for severe limb ischemia and it serves to promote angiogenesis and avoid amputation. However,our previous clinical practice and basic research found that diabetes suppressed angiogenesis of EPCs transplantation. There are many genes and factors involved in the course of EPCs mobilization, migration, differentiation and angiogenesis. It's unclear how diabetes suppress the course. The purpose of this research is to demonstrate the role of advanced glycation end products and its receptors (AGE-RAGE) in the suppression of EPCs angiogenesis in diabetic foot. In vivo and in vitro experiments will be conducted to illustrate the role of AGE-RAGE and its signal pathway in angiogenesis of ECPs transplantation. We hope that this research will help us get more understanding of the mechanism of EPCs transplantation and angiogenesis and thus improve the therapeutic effect of diabetic foot.
