中文摘要: microRNA是真核生物中一大类参与基因转录后水平调控的非编码小分子RNA,它们在生物体的细胞增殖、细胞分化、细胞凋亡、形态建成、个体发育、激素分泌与信号转导等一系列生命活动中起重要作用。过氧化物酶体对人的生长和发育非常重要,其合成缺乏会导致神经系统疾病。本项目以小鼠神经干细胞为材料,研究microRNA对过氧化物酶体生物合成和增殖的调控作用,发现miR-15、miR-16、miR-19、miR-27和miR-29能够调控过氧化物酶体生物合成和增殖相关基因的表达,通过分析这些microRNA及其所调控的靶基因的表达变化,解析了参与过氧化物酶体生物合成和增值的microRNA调控网络。本项目不仅阐明了microRNA在过氧化物酶体生物合成和增殖中的作用和调控机制,而且为探索与过氧化物酶体代谢相关的人类神经疾病的治疗方法提供了新思路。

英文摘要: microRNA(miRNA), a class of small non-coding RNAs that bind complementary mRNAs to regulate gene expression at the posttranscriptional level, are known to play crucial roles in many different cellular processes including metabolism, proliferation, differentiation and development, and defect in peroxisome biogenesis will result in nervous system diseases. In this study we investigated microRNAs which involved in the peroxisome biogenesis and proliferation and found that miR-15, miR-16, miR-27 and miR-29 can regulate expression of genes involved in peroxisome biogenesis and proliferation. We inferred the miRNA regulatory network regulating peroxisome biogenesis and proliferation by integrating expression levels of miRNAs with their potential mRNA targets. This study can not only illuminate the importance of microRNAs in peroxisome biogenesis and proliferation, but also provided a new strategy for exploring therapies for the human nervous system diseases related with peroxisome metabolism.
