中文摘要: 水凝胶是目前软组织填充材料产品的研究热点之一,软组织填充材料的生物学功效取决于材料加工过程和材料本身。不同的加工过程导致材料的内部结构差异很大,从而引起力学性能,降解性能和生物学功效的变化。通过化学共混和刚性分子与柔性分子对接的方法改变水凝胶的内部结构以及体系的构想变化,从而引起交联度、吸水率、弹性模量、孔径分别、孔隙率以及孔的贯通性的变化。本研究旨在阐释水凝胶在构建过程中工艺参数即不同分子量的普鲁兰多糖以及衍生物,类人胶原蛋白溶液的不同配比、粘度、空间连段结构、pH、温度等与材料的物理化学特性、表面形貌、力学性能、降解行为、生物学功效以及细胞粘附和增值的关系,解释刚性分子和柔性分子对接与结构构象变化的相关性;水凝胶的内部空间结构对水凝胶力学强度、降解行为以及组织修复的相关性;阐释接枝后的水凝胶对软组织填充和修复过程的贡献。为工业化生产过程中用于软组织填充及组织修复提供理论依据。

英文摘要: Currently, hydrogels are one of the hot products of soft tissue filler material. The biological efficacy of soft tissue filler material mainly depends on the processing and the composition of material. Different processing would result in the significant change of structural of the hydrogels, of course, leading to the big difference on the mechanical properties, degradation and biological efficacy. Changing the conformational and the internal structure of the hydrogel by a chemical blending system and the docking methods of rigid and flexible molecular, of course, cause the difference on the cross-linking degree, swelling ratio, the elastic modulus, pore size distribution, the porosity and pore changes . The target is to reveal to the correlation between the process parameters (different proportions, viscosity, even the space segment structure, pH, temperature during the processing between pullulan with different molecular weights and human-like collagen) and materials physical, chemical properties, surface morphology, mechanical properties, degradation behavior, biology, and cell adhesion, proliferation. To interpretation the correlation of the rigid and flexible molecular docking and the change of structure conformational; the relationship between internal space structure of the hydrogel and mechaical strength, degradation behavior and tissue repair, the contribution of grafted hydrogels to soft tissue filler and repair process。This study would give a theoretical guidance and basis on the soft tissue filler and repair to reach a better clinical application.
