中文摘要: 乳腺癌是女性最常见的恶性肿瘤,严重威胁女性生命健康。乳腺癌个体化医疗策略的实施,依赖于对其生物学行为的准确判断和评估。然而,乳腺癌异质性导致其生物学行为复杂,也制约其个体化医疗的实现。因此,深化对乳腺癌异质性研究,特别是构建癌细胞及微环境的多组分、多分子、原位共成像平台体系,是深入理解其异质性,制定个体化医疗策略的重要途径。新型纳米量子点(Quantum dots,QDs)标记分子探针影像技术在肿瘤分子原位、准确、灵敏、定量,多组分成像研究中具有显著优势,将该技术用于乳腺癌异质性研究,有望推动个体化医疗。本课题拟在既往基础上,充分发挥量子点标记分子探针影像技术及多学科交叉优势,通过对乳腺癌及其间质微环境组分及关键分子的原位、实时、定量、多组分、长时间分子成像,揭示癌细胞及其微环境异质性新特点,深化对乳腺癌生物学行为的理解,为制定个体化医疗策略提供理论和实践依据。

英文摘要: Breast cancer is the most common malignancy of women and a serious healthy threatens and burden to women. Personalized medicine strategy for each breast cancer relies on accurate and comprehensive evaluation its biological behavior. However, the heterogeneity of breast cancer makes the complexity of biological behaviors, and restricts the realization of personalized medicine. Therefore, deepen understanding the heterogeneity of breast cancer, especially for the establishment of new platform for multicellular, multi-molecule imaging of cancer cells and microenvironment is one of the important ways to personalized medicine strategy. Quantum dots (QDs) labeled molecular probes for the real-time, in situ, and co-location molecular imaging study and multi-spectral analysis technology have promising advantages in cancer research, especially in the field of breast cancer heterogeneity study. Based on this technology, previous multidisciplinary cooperation and results, this project aims at real-time, in situ, accurate, sensitive and quantitative imaging of key molecules in both cancer cells and their micro-environment in breast cancer cell lines, animal models and clinical specimens, and together with multi-spectral analyses and new algorithm, to reveal the new insights of breast cancer heterogeneity. With these new insights, we could deepen understanding the biological behavior of breast cancer, and help predict breast cancer invasive potential and formulate individualized treatment strategy.
