中文摘要: 脊髓损伤(SCI)修复机制研究与治疗是医学界关注的热点和难点。针刺促进损伤脊髓功能部分恢复已在临床得到证实,但分子机制不完全清楚。此前,我们通过RNA干扰及神经行为学评价探讨了SCI后脊髓内突触小体相关蛋白25(SNAP25)的作用及影像学表现 (Wang TH , Molecular Neurobiology, 2014, PubMed. PMID: 24519330)。进而,用蛋白组、Q-PCR、WB发现SCI后针刺能促进皮质内SNAP25表达上调。在此基础上,本项目拟建立大鼠脊髓横断伤督脉针刺模型,采用核磁共振成像、基因克隆、RNA干扰、免疫组织化学、Q-PCR、WB和细胞培养等技术,研究针刺促进SCI后皮质远程调控中SNAP25的作用及分子影像学特征。项目研究对推动针刺治疗SCI的临床应用有着重要的科学意义和临床价值,是具原创性和创新性强的研究工作。

英文摘要: The study on the mechanism and treatment after spinal cord injury (SCI) has been becoming the hotspot, and is attractive scientists to focus on SCI, a clinic severe problem in medical scientific field. It has been well known that acupuncture can promote functional recovery after SCI in clinical practice, but the underlying molecular mechanism was not fully understood. Our previous studies have investigated role of SNAP25 in the injured spinal cord of rats has been determined in our recent work, by using RNA interference and behavioral assessment (Wang TH *, Molecular Neurobiology, 2014, PubMed. PMID: 24519330); then found synaptosome-associated protein 25 (SNAP25) was specifically unregulated in the cortical and spinal cord of rats following SCI with acupuncture, by using two-dimensional electrophoresis; and this change was validated by Q-PCR and Western Blot. Moreover, in this study, we continuously explore the role of SNAP25 in remote cortex after SCI with acupuncture by using MRI, gene cloning, RNA interference, immunocytochemistry, Q-PCR, Western Blot and cell culture techniques. The findings should give more informations for the usage of acupuncture in clinical practice, which has important scientific implication and clinical meaning.
