中文摘要: 细胞是生命活动的基本单位,细胞内的能量代谢与细胞膜相行为研究对揭示生命的过程具有重要意义。已有的工作表明,单个细胞的温度会在外界刺激的作用下发生显著的变化。目前的脂筏模型阐释了鞘脂、胆固醇和特定的蛋白对细胞膜相行为以及区域划分的作用,但忽略了细胞自身代谢导致的温度变化对膜相行为及分区的影响或调控。 本课题拟利用金纳米双锥的近红外吸收性质,将其作为细胞内热源,并通过微纳热电偶探测细胞局部温度变化,以及共聚焦显微镜观测荧光标记的细胞膜相行为,建立局部温度变化与细胞膜相行为变化直接的关系。通过研究细胞膜的相行为与细胞局部温度变化之间的关系,为进一步理解细胞本身能量代谢水平的对细胞膜相行为的影响提供相关的证据。

英文摘要: Cell is the basic unit of living organisms. It is of great significance to investigate intracellular energy metabolism and cell membrane phase behavior for revealing the life process of cells. Literature and our previous work shows that, the temperature of the single cell change significantly by the specific stimulation. Lipid rafts model illustrated the sphingolipids, cholesterol, and specific membrane proteins that contribute to lateral heterogeneity in eukaryotic membranes. However, the temperature change by the cell metabolism which may influence membrane phase behavior was ignored. Whether the endogenous temperature change participate in controlling the membrane phase behavie or not, and how it works remain a question. In this research, to simplify the model, we propose to use gold nano cone with near infrared absorption properties, as the internal heat source, to detect local temperature change by the micro-thermocouple, to observe the fluorescence change which stand for membrane phase behavior by the confocal microscope. By studying the relationship between phase behavior of the cell membrane and the local temperature changes, it is expected to provide relevant evidence for the further understanding on how the cell of energy metabolism level influence to the cell membrane phase behavior.
