中文摘要: DNA的凝聚与解聚在基础生命科学、高分子物理以及应用医学等领域有重要意义。聚乙二醇能够导致DNA发生从线团构象到凝聚球状等构象的转变。由于聚乙二醇的无毒,无抗原等特性,其在生物医学领域有广泛应用和潜在价值。本课题拟研究聚乙二醇作用下的DNA的构象转变。课题拟利用磁镊、单分子力谱方法对DNA的构象转变过程在单分子水平上进行定量的实时动态研究,利用原子力显微镜对DNA在聚乙二醇作用下形成的构象进行扫描成像,利用动态光散射以及zeta电位测量技术研究DNA的尺寸分布和电位大小受聚乙二醇的影响。课题致力于明确DNA该体系中的构象及内部结构信息,构象转变的动力学机制,以及各种构象怎样在调控因素下转化,为从物理本质上理解构象转变机制以及为聚乙二醇在基因传递上的医学应用提供实验和模型参考。

英文摘要: DNA compaction and decompaction are important processes in life science, polymer physics and medical application. Polyethylene glycol(PEG) can cause the coil-globule transition of DNA. Because of its non-antigenicity and non-toxicity, it has great potential and has been used in biomedical field. This project intends the research on this transition. We plan to : 1)reveal the transition at a single molecule level using magnetic tweezer and single molecule force spectroscopy in order to clarify its dynamics in real time; 2)characerize the morphology of PEG-induced DNA structures using atomic force microscope; 3) measure the size distribution and zeta potential of DNA under the influence of PEG by dynamic light scattering and zeta potential methods. We aim to clarify the DNA conformation and its internal structure in this system, the dynamics of the transition how the conformation changes under modulating factors. This reseach can provide experimental and model references for the understanding of the transition mechanism and the its medical uses in gene delivery.
