中文摘要: 肺癌是目前最常见的恶性肿瘤之一,发病率和死亡率常年居高不下。肿瘤转移是肺癌病人死亡率高、预后极差的主要原因。本课题旨在利用microRNA表达谱芯片结合经典的分子生物学方法从非小细胞肺癌病人的临床样本中筛选与转移相关的microRNA,结合术后病人随访资料利用生物信息学方法,建立能够推断肺癌病人预后的特征性microRNA表达模型并完成验证。同时,通过细胞生物学和肿瘤行为学研究找出调控肺癌转移的重要功能microRNA,探明其生物学作用的分子机制并探讨其作为肺癌治疗靶点的可行性。前期工作中,我们利用microRNA芯片技术结合生物信息学分析对肺癌原发灶和转移灶中差异表达的microRNA进行了分析,已初步发现了一些可能与肺癌转移和预后相关的microRNA。本项目的顺利完成可望为肺癌转移检验及病人预后判定提供新的靶标。

英文摘要: Lung cancer is one of the most common malignancies with increasing incidence and mortality, due to invasive metastasis of tumor cells. This project utilizes microRNA array combined with classic approach of molecular biology to screen metastasis-related microRNA in tissue samples of patients with non-small cell lung cancer. Predictive microRNA signatures are established based on bioinformatic analysis of patients' prognosis. Further investigation will be focused on key microRNAs meditating metastasis of lung cancer, the underlying molecular mechanism and microRNAs as therapeutic targets of lung cancer treatment. Previous studies of microRNA array demonstrated that microRNAs differentially expressed in primary and metastatic lesions of lung cancer may be involved in the metastasis progress of lung cancer and correlated with prognosis. This study will provide new targets for diagnosis and prognosis of lung cancer.
